That'd be a drop of water, that you've dragged onto a clean surface, and dip the end of the applicator into a part of that smudge, just to barely moisten the cotton. Insert the tip of the applicator into the center of the port, apply a tiny bit of pressure, and then quickly snap it back out again. Wait a few minutes for that to dry, and then use compressed air. The point is to loosen the dirt with the applicator, so that the compressed air can blow it out.

Alcohol is not corrosive. Completely harmless, it's used to clean electronic parts. I invite anyone to look it up.
Compressed air may blow dirt deeper into your phone.
