Would be nice to have a dedicated room
I also use the guest room as my sewing room. Our 3rd bedroom doesn't have a good view for my sewing desk, and isn't as convenient to the guest bath, so we use that as an office/storage and to put our airbed in when we have guests in both rooms.
We have guests often enough that I don't want to ditch the queen bed in the guest room, but I have to admit I hate losing my sewing room the whole time they are here. I have thought about switching, but the 3rd bedroom is not convenient to the guest bath, and I love the front window for my sewing machine, plus it is a brighter room.
With some guests (our kids), I don't have time to sew, but with the others, it limits my early morning sewing time. This year, I'm going to move my sewing desk, machine and lamp, and a couple of projects, to the office when certain guests are anticipated so I don't lose out on my project time. Remains to be seen how that will work.
I'm getting to the point (after many guests) that I may not care how convenient their room is to the guest bath, but a queen bed, night stand and triple dresser might not fit in the 3rd bedroom.
I have seen closet set-ups, and it works for some, but I hate facing a wall when I am sewing so don't think I would like it. You are wise to think about this ahead of time.
